I know you can add to an existing audio recording. Take a look at this introduction video about the RealBand sequencer that comes with Band in a box. About 12 minutes in, it shows an example of adding generated instruments to an existing WMA file. MP3 should work just as well. If you want to create your own track to add, I'm sure Band in a Box could allow you to create it. There are a number of ways you can enter the notes. It's going to be interesting, there is a lot of capability in this product.

Re: Band-in-a-Box 2010This is really a very hasty guide. There are just too many options to explain. But see where this gets you. This is importing an MP3 or other audio file and generating chords, melody, style.
Select File > Open Audio w/Chords (WAV, WMA, MP3...) Select your audio file and click OK. Wait for the Audio Chord Wizard (ACW) dialog to open. Click the Video Help button to learn how to set Bar One and tap the barline, if needed. After making changes, click "OK - Send to BIAB" Click "Set the song key signature to -->" button. Click "Make a tempo map in BB (to sync MIDI with audio)" button. Check "Enable MIDI style." Click "Close" button. Click the green "Play" button to see what you've got so far. You can mute or solo the audio portion by clicking on the yellow text "Audio" near the top right and selecting what you want. To add a melody: From the menu go Melody > Melodist - Generate Melody and Chords, and clear the checkmark next to "Generate Chords" and "Allow Style Change." Then select a Genre and style, then click OK. You'll probably want to mute the audio at this point, so click "Audio" and select Mute from the pop-up window. Click the Play button to see what you've got. To change just the style, click the "Style..." button on the left side of the main window. Pick a catagory from the list on the left side and a style from the list in the middle. Click OK. To change an instrument -- Bass, Piano, Drums, Guitar, Strings, Melody, Soloist -- select the corresponding radio button at the top. Then in the next row of controls, select a new instrument from the dropdown list. The radio buttons that will be available to select depends on the style chosen.
